Opis: Recognizes a protein of 185 kDa, which is identified as c-erbB-2/HER-2/neu. Its epitope is localized in the extracellular domain. C-erbB-2/HER-2 is a member of the EGFR family. This MAb is specific and shows minimal cross-reaction with other members of the EGFR-family. Receptors of this family are located on the plasma membrane and consist of an extracellular ligand-binding domain that is connected to a large intracellular domain by a single transmembrane sequence. C-erbB-2/HER-2 protein is over-expressed in a variety of carcinomas especially those of breast and ovary.

Opis: This MAb reacts with the beta-chain of HLA-DRB1 antigen, a member of MHC class II molecules. It does not cross react with HLA-DP and HLA-DQ. HLA-DR is a heterodimeric cell surface glycoprotein comprised of a 36 kDa alpha (heavy) chain and a 28 kDa beta (light) chain. It is expressed on B-cells, activated T-cells, monocytes/macrophages, dendritic cells and other non-professional APCs. In conjunction with the CD3/TCR complex and CD4 molecules, HLA-DR is critical for efficient peptide presentation to CD4 T cells. It is an excellent histiocytic marker in paraffin sections producing intense cytoplasmic staining. True histiocytic neoplasms are similarly positive. HLA-DR antigens also occur on a variety of epithelial cells and their corresponding neoplastic counterparts. Loss of HLA-DR expression is related to tumor microenvironment and predicts adverse outcome in diffuse large B-cell lymphoma.

Opis: Recognizes a non-polymorphic determinant of DP-MHC class II. MHC class II antigens are transmembrane glycoproteins of non-covalently linked α (33-35 kDa) and β (27-30 kDa) chains. It reportedly reacts with B- & non-T, non-B cell lines but not with T- and myeloid cell lines and leukemias. Differential expression of MHC class II antigens on fetal and adult lymphocytes, malignant B cells appears to reflect the stage of cell differentiation which may be useful in the study of lymphoproliferative disorders.

Opis: This antibody recognizes a protein doublet of 20-22 kDa, identified as MART-1 (Melanoma Antigen Recognized by T cells 1) or Melan-A. MART-1 is a newly identified melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. Seven other melanoma associated antigens recognized by autologous cytotoxic T cells include MAGE-1, MAGE-3, tyrosinase, gp100, gp75, BAGE-1, and GAGE-1. Subcellular fractionation shows that MART-1 is present in melanosomes and endoplasmic reticulum. This MAb cocktail labels melanomas and other tumors showing melanocytic differentiation. It is also a useful positive-marker for angiomyolipomas. It does not stain tumor cells of epithelial, lymphoid, glial, or mesenchymal origin.

Opis: This antibody recognizes a ~90-95 kDa protein which is identified as cell surface transferrin receptor (CD71), a disulfide-bonded homodimeric glycoprotein of 180-190 kDa (Workshop IV). This MAb is highly specific to CD71 and shows no cross-reaction with other related proteins. Ligand for transferrin receptor is the serum iron transport protein, transferrin. This receptor is broadly distributed in carcinomas, sarcomas, leukemias, and lymphomas. CD71/Transferrin receptor has been reported to be associated with cell proliferation in both normal and neoplastic tissues and useful in predicting clinical behavior or response to therapy in a number of malignancies including breast cancer.

Opis: This antibody recognizes proteins of 80-200 kDa, identified as different members of CEA family. CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This MAb does not react with nonspecific cross-reacting antigen (NCA) and with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. Antibody to CEA is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are CEA ) from pleural mesotheliomas (rarely or weakly CEA ). Anti-CEA positivity is seen in adenocarcinomas from the lung, colon, stomach, esophagus, pancreas, gallbadder, urachus, salivary gland, ovary, and endocervix.

The Cdc2 protein kinase (p34Cdc2) exhibits protein kinase activity in vitro and exists in a complex with both cyclin B and a protein homologous to p13SUC1. Cdc2 kinase is the active subunit of the M phase promoting factor (MPF) and the M phase-specific Histone H1 kinase. The p34Cdc2/cyclin B complex is required for the G2 to M transition. An additional cell cycle-dependent protein kinase, termed p55cdc, exhibits a high degree of homology with the S. cerevisiae proteins Cdc20 and Cdc4. The p55cdc transcript is readily detectable in a variety of cultured cell lines in growth phase, but disappears when cell growth is chemically arrested.

Opis: This MAb reacts with a wide variety of simple epithelia. It does not react with stratified squamous epithelia. It reacts with epithelial tumors of the gastrointestinal tract, lung, breast, pancreas, ovary, and thyroid. Cytokeratin 18, which belongs to the type A (acidic) subfamily of low molecular weight keratins, exists in combination with cytokeratin 8. It was reported that tissues from gastrointestinal tract are positive for both cytokeratin 8 and 18 but do not contain cytokeratin 14. Tissues from gastrointestinal tract, respiratory tract and urogenital tract, as well as endocrine and exocrine tissues and mesothelial cells are positive for cytokeratin 18.

Opis: This MAb recognizes basic (Type II or HMW) cytokeratins, which include 67 kDa (CK1); 64 kDa (CK3); 59 kDa (CK4); 58 kDa (CK5); 56 kDa (CK6); 52 kDa (CK8). Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI 6.0) subfamilies. The acidic keratins have molecular weights (MW) of 56.5, 55, 51, 50, 50 , 48, 46, 45, and 40 kDa. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis.

Opis: This MAb reacts with the rod domain of human cytokeratin-19 (CK19), a polypeptide of 40 kDa. Its epitope maps between amino acid 312-335. CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. Anti-CK19 is a useful marker for detection of tumor cells in lymph nodes, peripheral blood, bone marrow and breast cancer.

Opis: Recognizes a glycoprotein of ~200 kDa, identified as carbonic anhydrase IX (CAIX/gp200). Its epitope resides in the carbohydrate domain of gp200. It shows no significant cross-reactivity with other carbohydrate determinants, such as the Lewis blood group antigens, epithelial membrane antigen, HMFG, and AB blood group antigens. In normal kidney, gp200 is localized along the brush border of the pars convoluta and pars recta segments of the proximal tubule, as well as focally along the luminal surface of Bowman's capsule adjoining the outgoing proximal tubule. Reportedly, gp200 is expressed by 93% of primary and 84% of metastatic renal cell carcinomas. This MAb may be useful in the investigations of carcinomas of proximal nephrogenic differentiation especially those showing tubular differentiation.
