Opis: Recognizes 183 kDa protein with DNA-binding characteristics, which is identified as a myeloid specific antigen. It reacts with myeloid precursor cells and granulocytes in bone marrow. Its antigen appears to be restricted to M2 and M3 acute myelogenous leukemia (AML) subtypes. Markers of myeloid cells are useful in the identification of different levels of cellular differentiation. This MAb reacts with early precursor and mature forms of human myeloid cells. It is useful in the identification of myelogenous leukemias, distinguishing granulocytic sarcomas from lymphoid malignancies and also in the study of differentiation and transformation of human myeloid cells. The biological function of this antigen is not clear, although it has been proposed that it may play a role in the differentiation of myeloid cells.

It can be used to stain the Golgi complex in cell or tissue preparations and can be used as a Golgi marker in subcellular fractions. It produces a diffuse staining pattern of the Golgi zone in normal and malignant cells. This MAb is an excellent marker for human cells in xenographic model research. It reacts specifically with human cells. The Golgi apparatus is an organelle present in all eukaryotic cells that forms a part of the endomembrane system. The primary function of the Golgi apparatus is to process and package macromolecules synthesized by the cell for exocytosis or use within the cell. The Golgi is made up of a stack of flattened, membrane-bound sacs known as cisternae, with three functional regions: the cis face, medial region and trans face. Opis: This MAb reacts with a reduction-resistant epitope present in both free and SIgA bound Secretory Component. It does not react with the cell lines lacking secretory component. The antibody is useful for studying the distribution and level of both free and bound secretory component. Secretory component is differentially expressed in epithelium, and the antibody is a popular marker for identifying subpopulations of epithelial cells and epithelial differentiation. Proinsulin, which has very little biological activity, is cleaved by proteases within its cell of origin into the insulin molecule and the C-terminal basic residue. Insulin enhances membrane transport of glucose, amino acids, and certain ions. It also promotes glycogen storage, formation of triglycerides, and synthesis of proteins and nucleic acids. Deficiency of insulin results in diabetes mellitus. The main storage site for insulin is the pancreatic islets. Antibodies to insulin are important as beta-cell and insulinoma marker.

Opis: This MAb reacts specifically with heat shock protein HSP27 in human and monkey tissues and cell lines such as MCF-7. HSP27, also referred to as the Estrogen-Regulated 24K protein and HSP28, is one of several small heat shock proteins produced by all organisms studied. HSP27 synthesis is induced by elevated temperature, as well as by estrogen in hormone responsive cells. Interestingly, human HSP27 also shares greater than 50% homology with low molecular weight Drosophila HSPs and mammalian α-crystalline lens protein. Because of the estrogen responsive nature of HSP27, this protein has been studied extensively in human estrogen responsive tissues such as cervix, endometrium and breast tissue. Therefore HSP27 may be useful in classifying various hormone sensitive tumors.

Opis: CA19-9, a carbohydrate epitope expressed on a high MW (>400 kDa) mucin glycoprotein, is a sialyl Lewisa structure which is synthesized from type 1 blood group precursor chains and is present in individuals expressing the Lewisa and/or Lewisb blood group antigens. In normal tissues, sialyl Lewisa antigen is present in ductal epithelium of the breast, kidney, salivary gland, and sweat glands. Its expression is greatly enhanced in serum as well as in the majority of tumor cells in gastrointestinal (GI) carcinomas, including adenocarcinomas of the stomach, intestine, and pancreas. Preoperative elevated CA19-9 levels in patients with stage I pancreatic carcinoma decrease to normal values following surgery. When used serially, CA19-9 can predict recurrence of disease prior to radiographic or clinical findings. This MAb is excellent for staining of formalin-fixed, paraffin-embedded tissues.

HMFG is present on normal human breast epithelial cells and cell lines derived from breast carcinomas, as well as to the outer surface of the human milk fat globule. HMFG is considered as a differentiation marker. It is useful as specific breast epithelial marker and can also provide a tool to study the role of the cell surface in normal and neoplastic mammary development.

Opis: This MAb is specific to kappa light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. It recognizes human Ig kappa light chains of both secreted and cell surface immunoglobulin. It detects also free kappa light chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.

Opis: This MAb recognizes a protein of 145 kDa, identified as CD117/p145kit. It is found on a wide variety of tumor cells including follicular and papillary carcinoma of thyroid, adenocarcinomas from endometrium, lung, ovary, pancreas, and breast as well as malignant melanoma, endodermal sinus tumor, and small cell carcinoma. However, anti-CD117 has been particularly useful in differentiating gastrointestinal stromal tumors from Kaposi's sarcoma, tumors of smooth muscle origin, fibromatosis, and neural tumors of the GI tract. Anti-CD117 is also useful in recognizing myeloblasts in bone marrow biopsy and clot section.
