Opis: This MAb preferably reacts with determinants of chain A and H type 3(Gal1-3GalNAc-R) and 4 (Gal1-3GalNAc-R), but not with type 1 and 2 chain structures. It is not reactive with immuno-dominant A trisaccharide. This MAb is applicable for tissue staining in tumor patients with blood groups A and AB. It shows a highly heterogeneous reactivity in human colon tumor tissue and adjacent mucosa. Blood-group antigens are generally defined as molecules formed by sequential addition of saccharides to the carbohydrate side chains of lipids and proteins detected on erythrocytes and certain epithelial cells. The A, B and H antigens are reported to undergo modulation during malignant cellular transformation. Blood group related antigens represent a group of carbohydrate determinants carried on both glycolipids and glycoproteins. They are usually mucin-type, and are detected on erythrocytes, certain epithelial cells, and in secretions of certain individuals. Sixteen genetically and biosynthetically distinct but inter-related specificities belong to this group of antigens, including A, B, H, Lewis A, Lewis B, Lewis X, Lewis Y, and precursor type 1 chain antigens.

Expression of bcl-2 α oncoprotein inhibits the programmed cell death (apoptosis). In most follicular lymphomas, neoplastic germinal centers express high levels of bcl-2 α protein, whereas the normal or hyperplastic germinal centers are negative. Consequently, this antibody is valuable when distinguishing between reactive and neoplastic follicular proliferation in lymph node biopsies. It may also be used in distinguishing between those follicular lymphomas that express bcl-2 protein and the small number in which the neoplastic cells are bcl-2 negative.

Grp94 shows a high degree of sequence homology with the heat shock protein 90 (hsp90). This MAb is highly specific to grp94 and shows minimal cross-reaction with other members of the HSP90 family. Grp s are a class of proteins unresponsive to heat shock and are induced by glucose deprivation. Grp94 has been briefly studied as a prognostic factor in breast cancer.

Opis: CD45R, also designated CD45 and PTPRC, has been identified as a transmembrane glycoprotein, broadly expressed among hematopoietic cells. Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. CD45R functions as a phosphor-tyrosine phosphatase. This MAb reacts with all isoforms of CD45R expressed by all hematopoietic cells, except erythrocytes, having a higher level of expression on lymphocytes than on granulocytes (Workshop IV). Antibody to CD45 is useful in differential diagnosis of lymphoid tumors from non-hematopoietic undifferentiated neoplasms.
