Opis: This antibody recognizes a protein of 30 kDa, identified as CD70. It is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This cytokine is a ligand for TNFRSF27/CD27. It is a surface antigen on activated, but not on resting, T- and B-lymphocytes. It induces proliferation of co-stimulated T cells, enhances the generation of cytolytic T cells, and contributes to T cell activation. This cytokine is also reported to play a role in regulating B-cell activation, cytotoxic function of natural killer cells, and immunoglobulin synthesis. This MAb blocks the interaction between CD27 and CD70, and has been shown toinhibit T cell proliferation induced by dendritic cells.

Opis: This MAb recognizes extracellular epitope of human CD147. It is expressed more intensely on thymocytes than on mature peripheral blood T cells. CD147 is important in spermatogenesis, embryo implantation, neural network formation, and tumor progression. It stimulates the production of interstitial collagenase, gelatinase A, stromelysin-1 and various metalloproteinases (MMPs) by fibroblasts. These enzymes are important factors in cancer invasion and metastasis.

Opis: This antibody recognizes a transmembrane glycoprotein of 95 kDa, identified as CD18 or integrin -2 (Workshop III). It complexes non-covalently with either L, M, or X integrin (CD11a, b, or c) to form the heterodimers, LFA-1, Mac-1, and p150,95, respectively. LFA-1 is the receptor for three members of the Ig supergene family of proteins, ICAM-1 (CD54), ICAM-2 (CD102), and Mac-1 and p150,95 bind to ICAM-1, fibrinogen, and iC3b. ICAM-3 (CD50). CD18/CD11 heterodimeric molecules are involved with cell/cell and cell/extracellular adhesion in immune and inflammatory responses. This MAb blocks these cellular interactions.

Opis: Smooth muscle myosin heavy chain (SM-MHC) is a cytoplasmic structural protein, which is a major component of the contractile apparatus in smooth muscle cells. Expression of smooth muscle myosin is developmentally regulated, appearing early in smooth muscle development, and is specific for smooth muscle development. Two isoforms of smooth muscle myosin heavy chain have been identified, designated MHC-1 and MHC-2. The antibody may be useful for the study of breast tumors as the presence of an intact layer of myoepithelial cells is an important feature, which may distinguish benign breast lesions and carcinoma in situ from invasive tumors.

CD86 is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily of cell surface receptors. It is expressed at high levels on resting peripheral monocytes and dendritic cells and at very low density on resting B and T lymphocytes. CD86 expression is rapidly upregulated by B cell specific stimuli with peak expression at 18 to 42 hours after stimulation. CD86, along with CD80/B71, is an important accessory molecule in T cell co-stimulation via its interaction with CD28 and CD152/CTLA4. Since CD86 has rapid kinetics of induction, it is believed to be the major CD28 ligand expressed early in the immune response. It is also found on malignant Hodgkin and Reed Sternberg (HRS) cells in Hodgkin's disease.

It is expressed on lymphocyte cells and is upregulated during T-cell activation. CD26 is also expressed on activated B cells and natural killer cells and abundantly on epithelia. CD26 is implicated in a variety of biological functions including T-cell activation, cell adhesion with extracellular matrix such as fibronectin or collagens, and in HIV infection. Cross-linking of CD26 using this antibody dramatically enhances the anti-CD3-induced IL-2 production. In Western blotting, this MAb reacts with only glycosylated CD26, but not with the deglycosylated form. It does not prevent ADA binding to CD26.

Opis: This antibody recognizes a glycoprotein of 110 kDa, which is identified as CD68. It is important for identifying macrophages in tissue sections. It stains macrophages in a wide variety of human tissues, including Kupffer cells and macrophages in the red pulp of the spleen, in lamina propria of the gut, in lung alveoli, and in bone marrow. It reacts with myeloid precursors and peripheral blood granulocytes. It also reacts with plasmacytoid T cells, which are supposed to be of monocyte/macrophage origin. It shows strong granular cytoplasmic staining of chronic and acute myeloid leukemia and also reacts with rare cases of true histiocytic neoplasia. Lymphomas are negative or show few granules.

Opis: von Willebrand Factor (vWF) is a multimeric glycoprotein that is found in endothelial cells, plasma and platelets. It acts as a carrier protein for Factor VIII and promotes platelet adhesion and aggregation. vWF undergoes a variety of posttranslational modifications that influence the affinity and availability for Factor VIII, including cleavage of the propeptide and formation of N-terminal disulfide bonds. This antibody helps to establish the endothelial nature of some lesions of disputed histogenesis, e.g. Kaposi's sarcoma and cardiac myxoma. It is widely used for differentiating vascular lesions from those of other tissue differentiation within a panel of other vascular markers although not all tumors of endothelial differentiation contain this antigen.

Opis: Recognizes a 47-55 kDa-tumor suppressor protein, identified as Wilm's Tumor (WT1) protein. The antibody reacts with all isoforms of the full-length WT1 and also identifies WT1 lacking exon 2-encoded amino acids, frequently found in subsets of sporadic Wilm s tumors.WT1, a sporadic and familial pediatric kidney tumor, is genetically heterogeneous. Wilm s tumor is associated with mutations of WT1, a zinc-finger transcription factor that is essential for the development of the metanephric kidney and the urogenital system. The WT1 gene is normally expressed in fetal kidney and mesothelium, and its expression has been suggested as a marker for Wilm s tumor and mesothelioma. WT1 protein has been identified in proliferative mesothelial cells, malignant mesothelioma, ovarian carcinoma, gonadoblastoma, nephroblastoma, and desmoplastic small round cell tumor. Lung adenocarcinomas rarely stain positive with this antibody. WT1 protein expression in mesothelial cells has become a reliable marker for the diagnosis of mesotheliomas.

ZAP70 protein is expressed in leukemic cells of approximately 25% of chronic lymphocytic leukemia (CLL) cases as well.Anti-ZAP70 expression is an excellent surrogate marker for the distinction between the Ig-mutated (anti-ZAP70 negative) and Ig-unmutated (anti-ZAP70 positive) CLL subtypes and can identify patient groups with divergent clinical courses. The anti-ZAP70 positive Ig-unmutated CLL cases have been shown to have a poorer prognosis.

Opis: This antibody recognizes a 66 kDa protein, which is identified as Tripartite motif-containing protein 29 (TRIM29). It interacts with the intermediate filament protein vimentin, a substrate for the PKC family of protein kinases, and with hPKCI-1, an inhibitor of the PKCs. TRIM29 protein contains both zinc finger and leucine zipper motifs, suggesting that the it may form homodimers and possibly associate with DNA. High expression of TRIM29 has been reported in gastric cancer and pancreatic cancer, and correlates with enhanced tumor growth and lymph node metastasis. TRIM29 is also able to distinguish lung squamous cell carcinoma from lung adenocarcinoma with ~90% positive accuracy, when used in a panel with TTF-1, p63, CK5/6, and Napsin-A antibodies.
