Opis: CD17 is an intermediate glycosphingolipid from the metabolism of higher gangliosides that localizes to sphingolipid-sterol rafts. CD17 is detectable in monocytes, granulocytes, basophils, platelets, a subset of peripheral B cells (CD19 ) and tonsil dendritic cells. It is rapidly down regulated on activated granulocytes and is upregulated on IL-2 activated T lymphocytes. CD17 binds to bacteria and may function in phagocytosis. VEGF-treated endothelial cells can produce CD17, which can then mediate signaling toward PECAM-1 expression and angiogenesis. Tumor necrosis factor alpha (TNFa)-induced astrogliosis (astrocyte proliferation and glial fibrillary acidic protein (GFAP) upregulation) in response to neuro-inflammation (i.e. spinal cord injury) causes an increase in intracellular levels of CD17. Aberrant levels of glycosphingolipids are a feature of cancer cells and may influence integrin clustering and internalization.

In some patients with AIDS, CMV is detected in the bronchoalveolar lavage fluid (BALF), urine, and other specimens, even when there are no symptoms of CMV disease. An indicator of active CMV infection is needed to facilitate the diagnosis of CMV disease in patients with AIDS or HIV infection. CMV p65 antigen was detected in the leukocytes of both the peripheral blood and BALF during the early phase of CMV disease.

Opis: This MAb reacts with the beta-chain of HLA-DRB1 antigen, a member of MHC class II molecules. It does not cross react with HLA-DP and HLA-DQ. Its epitope is different from that of MAb L243. HLA-DR is a heterodimeric cell surface glycoprotein comprised of a 36 kDa alpha (heavy) chain and a 28 kDa beta (light) chain. It is expressed on B-cells, activated T-cells, monocytes/macrophages, dendritic cells and other non-professional APCs. In conjunction with the CD3/TCR complex and CD4 molecules, HLA-DR is critical for efficient peptide presentation to CD4 T cells. It is an excellent histiocytic marker in paraffin sections producing intense cytoplasmic staining. True histiocytic neoplasms are similarly positive. HLA-DR antigens also occur on a variety of epithelial cells and their corresponding neoplastic counterparts. Loss of HLA-DR expression is related to tumor microenvironment and predicts adverse outcome in diffuse large B-cell lymphoma.

CD19 is present in both benign and malignant B-cells and is considered to be the most reliable surface marker of this lineage over a wide range of maturational stages. In normal lymphoid tissue, CD19 is observed in germinal centers, in mantle zone cells, and in scattered cells of the inter-follicular areas. Anti-CD19 exhibits an overall immunoreactivity pattern similar to those of the antibodies against CD20 and CD22. However, in contrast to CD20, expression of CD19 is continuous throughout B-cell development and through terminal differentiation of B-cells into plasma cells. Anti-CD19 positivity is seen in the vast majority of B-cell neoplasms commonly at a lower intensity than normal B-cell counterparts. Plasma cell neoplasms are nearly always negative, as are T-cell neoplasms.

Opis: Recognizes a protein of 30-33 kDa, which is identified as CD20 (Workshop V; Code CD20.4). It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. The protein passes through the membrane 4 times with both ends in cytoplasm and exposes one short and one longer loop to the external environment. CD20 is not glycosylated in resting B-cells and its cytoplasmic domains are differentially phosphorylated upon activation. It acts as calcium channel involved in B cell activation and cell cycle progression.

CD11a complex with the 2 subunit of the integrin family, CD18, to form the cell surface heterodimer, LFA-1 or CD11a /C18 (aLbL). LFA-1 is expressed on all leukocytes including lymphocytes, monocytes, and granulocytes. It is involved in leukocyte adhesion to its ligands including intercellular adhesion molecule-1 (ICAM-1 or CD54), ICAM-2 (CD102), ICAM-3 (CD50) and Telencephalin (TLN) and play a role in most immune/inflammatory responses. This MAb potently blocks LFA-1 dependent homotypic cell aggregation.

CD38 is a type II transmembrane glycoprotein that is present on early B- and T-cell lineages and activated B- and T-cells but is absent from most mature resting peripheral lymphocytes. CD38 is also found on thymocytes, pre-B cells, germinal center B-cells, mitogen-activated T-cells, monocytes and Ig-secreting plasma cells. CD38 is expressed on CD34 cells. The CD34 CD38- population of hematopoietic stems cells defines the most pluripotent cells (e.g. blast colony forming cells).

Opis: Recognizes a cell surface glycoprotein of 80-95 kDa (CD44) on lymphocytes, monocytes, and granulocytes (Leucocyte Typing Workshop V). Its epitope is resistant to digestion by trypsin and chymotrypsin. The CD44 family of glycoproteins exists in a number of variant isoforms, the most common being the standard 85-95 kDa or hematopoietic variant (CD44s). Higher molecular weight isoforms are described in epithelial cells (CD44v), which are believed to function in intercellular adhesion and stromal binding. CD44 immunostaining is commonly used for the discrimination of urothelial transitional cell carcinoma in-situ from non-neoplastic changes in the urothelium.
