Opis: This product is prepared by labeling highly cross-adsorbed donkey anti-rat IgG (H+L) with CF™405S. CF™405S is a blue fluorescent dye with an absorption peak wavelength that nearly coincides with the 405 nm blue diode laser line. In addition, the emission peak wavelength of the dye well centers within the blue detection window of BD flow cytometers. As a result, flow cytometry analysis using CF™405S dye results in much brighter signal than that using Alexa Fluor® 405.   To minimize cross-reactivity, the antibody has been adsorbed against bovine, chicken, goat, guinea pig, Syrian hamster, horse, human, mouse, rabbit, and sheep serum proteins .

Opis: Glypican-3 (GPC3) is a glycosylphospatidyl inositol-anchored membrane protein, which may also be found in a secreted form. Anti-GPC3 has been identified as a useful tumor marker for the diagnosis of hepatocellular carcinoma (HCC), hepatoblastoma, melanoma, testicular germ cell tumors, and Wilm s tumor. In patients with HCC, GPC3 is overexpressed in neoplastic liver tissue and elevated in serum, but is undetectable in normal liver, benign liver, and the serum of healthy donors. GPC3 expression is also found to be higher in HCC liver tissue than in cirrhotic liver or liver with focal lesions such as dysplastic nodules and areas of hepatic adenoma (HA) with malignant transformation. In the context of testicular germ cell tumors, GPC3 expression is up regulated in certain histologic subtypes, specifically yolk sac tumors and choriocarcinoma. A high level of GPC3 expression is also found in some types of embryonal tumors, such as Wilm s tumor and hepatoblastoma, with a low or undetectable expression in normal adjacent tissue. In patients with thyroid cancer, expression of GPC3 is dramatically enhanced in certain types of cancers: 100% in follicular carcinoma and 70% in papillary carcinoma. Expression of GPC3 in follicular carcinoma is significantly higher than that of follicular adenoma. In contrast, GPC3 is not expressed in anaplastic carcinoma.

Opis: Recognizes a protein of 47-55 kDa, which is identified as FOXP3. Its precise epitope is not known, but it has been mapped to the N-terminal portion of the protein. The FOX family of transcription factors is a large group of proteins that share a common DNA binding domain termed a winged-helix or forkhead domain. During early development, FOXP1 and FOXP2 are expressed abundantly in the lung, with lower levels of expression in neural, intestinal and cardiovascular tissues, where they act as transcription repressors. FOXP1 is widely expressed in adult tissues, while neoplastic cells often exhibit a dramatic change in expression level or localization of FOXP1. Mutations in FOXP3 gene cause IPEX, a fatal, X-linked inherited disorder characterized by immune dysregulation. The FOXP3 protein is essential for normal immune homeostasis. Specifically, FOXP3 represses transcription through a DNA binding forkhead domain, thereby regulating T cell activation.

Opis: Recognizes a non-polymorphic determinant of DP-MHC class II. MHC class II antigens are transmembrane glycoproteins of non-covalently linked α (33-35 kDa) and β (27-30 kDa) chains. It reportedly reacts with B- & non-T, non-B cell lines but not with T- and myeloid cell lines and leukemias. Differential expression of MHC class II antigens on fetal and adult lymphocytes, malignant B cells appears to reflect the stage of cell differentiation which may be useful in the study of lymphoproliferative disorders.

Human MHC class II antigens are transmembrane glycoproteins composed of an chain (36 kDa) and a chain (27 kDa). They are expressed primarily on antigen presenting cells such as B lymphocytes, monocytes, macrophages, and thymic epithelial cells and are also present on activated T lymphocytes. Human MHC class II genes are located in the HLA-D region that encodes at least six and ten chain genes. Three loci, DR, DQ and DP, encode the major expressed products of the human class II region. The human MHC class II molecules bind intracellularly processed peptides and present them to T-helper cells. They, therefore, have a critical role in the initiation of the immune response. It has been shown that some autoimmune diseases are associated with certain class II alleles.

Opis: This MAb reacts with the beta-chain of HLA-DRB1 antigen, a member of MHC class II molecules. It does not cross react with HLA-DP and HLA-DQ. HLA-DR is a heterodimeric cell surface glycoprotein comprised of a 36 kDa alpha (heavy) chain and a 28 kDa beta (light) chain. It is expressed on B-cells, activated T-cells, monocytes/macrophages, dendritic cells and other non-professional APCs. In conjunction with the CD3/TCR complex and CD4 molecules, HLA-DR is critical for efficient peptide presentation to CD4 T cells. It is an excellent histiocytic marker in paraffin sections producing intense cytoplasmic staining. True histiocytic neoplasms are similarly positive. Integrins are heterodimeric proteins that contain an a chain and b chain. Integrin αM combines with the Integrin β2 to form a leukocyte-specific integrin referred to as macrophage receptor 1 (Mac-1), or inactivated-C3b (iC3b) receptor 3 (CR3). Integrin αM/β2 is important in the adherence of neutrophils and monocytes to stimulated endothelium, and also in the phagocytosis of complement coated particles.
