Opis: Cyclic AMP-dependent phosphodiesterase type D (PDE4D) family is comprise of 5 variants (PDE4D1, D2, D3, D4 and D5). One or more PDE4D subtype variants are ubiquitously present in all mammalian cells. In CNS all five PDE4D subtype variants are expressed in varying ratios and their activity is regulated in tandem with GPCRs stimulation. Peripheral tissues also exhibit differential expression of PDE4D variants. PDE4D1/D2 mRNA levels rise in response to an increase in cAMP. Short term regulation of PDE4D variants involved PKA, MAP kinases and Erk2 phosphorylation that results in rapid change in their enzymatic activities. Other regulatory mechanism involved protein protein interactions with cytoskeletal scaffolding proteins.

Opis: Receptor tyrosine kinase involved in the development and the maturation of the central and the peripheral nervous systems through regulation of neuron survival, proliferation, migration, differentiation, and synapse formation and plasticity. Receptor for BDNF/brain-derived neurotrophic factor and NTF4/neurotrophin-4. Alternatively can also bind NTF3/neurotrophin-3 which is less efficient in activating the receptor but regulates neuron survival through NTRK2. Upon ligand-binding, undergoes homodimerization, autophosphorylation and activation. Recruits, phosphorylates and/or activates several downstream effectors including SHC1, FRS2, SH2B1, SH2B2 and PLCG1 that regulate distinct overlapping signaling cascades. Through SHC1, FRS2, SH2B1, SH2B2 activates the GRB2-Ras-MAPK cascade that regulates for instance neuronal differentiation including neurite outgrowth. Through the same effectors controls the Ras-PI3 kinase-AKT1 signaling cascade that mainly regulates growth and survival. Through PLCG1 and the downstream protein kinase C-regulated pathways controls synaptic plasticity. Thereby, plays a role in learning and memory by regulating both short term synaptic function and long-term potentiation. PLCG1 also leads to NF-Kappa-B activation and the transcription of genes involved in cell survival. Hence, it is able to suppress anoikis, the apoptosis resulting from loss of cell-matrix interactions. May also play a role in neutrophin-dependent calcium signaling in glial cells and mediate communication between neurons and glia.

Opis: Aurora related kinase-1 (ARK-1) is a centrosome-associated serine/threonine kinase that regulates centrosome separation, bipolar spindle assembly and chromosome segregation during mitosis. Bora (protein aurora borealis) is a 559 amino acid protein that activates ARK-1. Bora is localized to the nucleus until mitosis is initiated, when it then translocates to the cytoplasm. This translocation is dependent on activated Cdc2, which releases Bora to bind and activate ARK-1 in the cytoplasm. Plk (polo-like kinase) interacts with Bora to control the accessibility of its activation loop for phosphorylation and activation on its N-terminus by ARK-1. It is through this mechanism that Bora and ARK-1 control cellular mitotic entry. Downregulation of the gene encoding Bora results in multipolar spindles in mitosis, a phenomenon that is also observed when ARK-1 function is blocked.

Opis: Ubiquitination is an important mechanism through which three classes of enzymes act in concert to target short-lived or abnormal proteins for destruction. The three classes of enzymes involved in ubiquitination are the ubiquitin-activating enzymes (E1s), the ubiquitin-conjugating enzymes (E2s) and the ubiquitin-protein ligases (E3s). UBC13, also known as UBE2N or BLU, is a 152 amino acid member of the E2 ubiquitin-conjugating enzyme family. Existing as a heterodimer with Mms2 (also known as UBE2V2), UBC13 catalyzes the ATP-dependent synthesis of non-canonical polyubiquitin chains, a process that does not lead to proteasomal degradation. Additionally, UBC13 mediates the transcription of several target genes and is thought to play a role in cell cycle progression, cellular differentiation and DNA repair mechanisms that ensure cell survival after DNA damage.

Opis: Estrogen receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of ligand-activated transcription factors. Estrogen receptors, including ER alpha and ER beta, contain DNA binding and ligand binding domains and are critically involved in regulating the normal function of reproductive tissues. ER alpha and ER beta A have been shown to be differentially activated by various ligands. Receptor-ligand interactions trigger a cascade of events, including dissociation from heat shock proteins, receptor dimerization, phosphorylation and the association of the hormone activated receptor with specific regulatory elements in target genes. Evidence suggests that ER alpha and ER beta may be regulated by distinct mechanisms even though they share many functional characteristics.

Opis: The Per-Arnt-Sim (PAS) domain is a 270 amino acid motif that mediates associations among various PAS family transcription factors. The PAS family contains neuronal specific transcription factors known as NPAS1, NPAS2 and NPAS3, which are involved the development and maintenance of learning and memory pathways. NPAS1 regulates erythropoietin expression in developing brain. NPAS2, also designated PAS 4/MOP4, associates with MOP3 to activate transcription. NPAS3, which localizes to the nucleus and is ubiquitously expressed in the adult brain, may be involved in neurogenesis and may control regulatory pathways relevant to psychotic illness and to schizophrenia. It regulates tracheal cell fates in the embryo and is necessary for the development of the posterior spiracles and the salivary gland duct. NPAS3 contains 1 basic helix-loop-helix (bHLH) domain, 1 PAC (PAS-associated C-terminal) domain, and 2 PAS (PER-ARNT-SIM) domains. Efficient DNA binding by NPAS2 requires dimerization with another bHLH protein.

Opis: A hands on lab activity to study immunoaffinity chromatography and use a specific antibody to purify antigenic proteins from complex samples. This technique involves performing a chromatography procedure using antigen or antibody immobilised on a chromatographic resin. The solution containing antigen or antibody is passed through the column, which specifically and efficiently captures antibodies (antigen). The captured molecules are eluted from the column as a pure fraction. In this lab activity, students perform immunoaffinity chromatography and learn how this method is utilised in research laboratories.

Opis: GALR3 a 368 and 370 amino acid protein in human and rat, respectively, belongs to a family of G protein-coupled receptors that bind the neuropeptide galanin, which is distributed throughout the central and peripheral nervous system, the pituitary gland, the gastrointestinal tract and in the endocrine and exocrine pancreas. GALR3 mRNA is widely distributed, but expressed at low abundance. In human, GALR3 mRNA is highly expressed in the hypothalamus, pituitary and testis, and is expressed to a lesser extent in adrenal gland and pancreas. Rat and human GALR3 co-express with potassium channel subunits GIRK1 and GIRK4. Like GALR1, GALR3 signaling pathways lead to the inhibition of adenylate cyclase and to the activation of potassium channels, which are linked to the regulation of neurotransmitter release. Binding of galanin to galanin receptors results in increased feeding, impaired learning, enhanced opiate analgesia and decreased opiate place preference.
